By bus you will need to take either Caribe Tours or Metro bus from POP to Santo Domingo. Then take Expresso Bavaro bus to Punta Cana. Search any of these companies for further explanation. It isn't a short trip. You can get Caribe and Metro schedules online. They are very regular in the morning. Departures from Santo Domingo on Expresso Bavaro are at 7 am, 10 am, 2 pm and 4 pm.

PS

Caribe only gives departure times in POP, no arrival times in SD.
How long approx. is the trip?

If memory serves me right, there is a 20 - 30 minute stop in Santiago, and the entire trip is 4 hrs.

A few minutes by taxi, they're on the same side of the capital but not quite walking distance.

Make sure to time your buses to arrive early enough for the Bavaro Express or you will be left there. You need to get there at least 30 minutes early as it does fill up! It is a very long trip. Take a jacket or sweatshirt as the buses are heavily air conditioned.

It is a long day bring a book and something to snack on.
